Potential of Microsecond Pulse Electric Fields (µsPEF) for Epigenetic Manipulation and Breast Cancer Treatment
In this study, we stimulated nucleocytoplasmic shuttling of HDAC4 in MCF7 cells by applying 300 V for 100 ms with varying numbers of square-wave pulses (1, 10, or 50) at 1 Hz repetition rate. We postulate that increased Ca2+ uptake due to msPEF exposure activated calmodulin and CaMKII, leading to HDAC4 translocation outside nuclei. Disruption of proliferation of MCF7 breast cancer cells was manipulated via msPEF exposure-induced inhibition of HDAC4.
