(721i) Effect of Pore Size and Particle Diameter on Adsorption and Elution Performance of Hydroxyapatite Chromatography

Effects of pore size and particle diameter on adsorption (dynamic binding capacity) and elution (peak salt concentration and peak width) in hydroxyapatite chromatography were investigated. The retention volume of proteins decreased with increasing pore size irrespective of particle diameter. The binding site value was not markedly affected by pore size. Dynamic binding capacity (DBC) of immunoglobulin G depended strongly on particle size and pore diameter. We established a new correlation between DBC and a normalized residence time with respect to particle diameter.
